Emb15 encodes a plastid ribosomal assembly factor essential for embryogenesis in maize

چکیده

Ribosome assembly factors guide the complex process by which ribosomal proteins and RNAs form a functional ribosome. However, of plant plastid ribosomes is poorly understood. In present study, we discovered maize (Zea mays) ribosome factor based on our characterization embryo defective 15 (emb15) mutant. Loss function Emb15 retards development at an early stage, but does not substantially affect endosperm, causes albino phenotype in other genetic backgrounds. EMB15 localizes to plastids possesses maturation M (RimM) domain N-terminus predicted UDP-GlcNAc pyrophosphorylase C-terminus. The RimM originated bacteria fungi; these two domains came together ancestor land plants during evolution. complemented growth defect Escherichia coli strain with deletion rescued emb15 homozygous mutants. mediates interaction between protein PRPS19. Plastid 16S rRNA also significantly impaired emb15. These observations suggest that functions seed as factor, C-terminal important under normal conditions.

Tobacco plastid ribosomal protein S18 is essential for cell survival

Plastid genomes contain a conserved set of genes most of which are involved in either photosynthesis or gene expression. Among the ribosomal protein genes present in higher plant plastid genomes, rps18 is special in that it is absent from the plastid genomes of several non-green unicellular organisms, including Euglena longa and Toxoplasma gondii. Here we have tested whether the ribosomal prote...

EMBRYONIC FACTOR 19 encodes a pentatricopeptide repeat protein that is essential for the initiation of zygotic embryogenesis in Arabidopsis.

Early embryogenesis is the most fundamental developmental process in biology. Screening of ethyl methanesulfonate (EMS)-mutagenized populations of Arabidopsis thaliana led to the identification of a zygote-lethal mutant embryonic factor 19 (fac19) in which embryo development was arrested at the elongated zygote to octant stage. The number of endosperm nuclei decreased significantly in fac19 emb...

A protein factor essential for microtubule assembly.

A heat stable protein essentail for microtubule assembly has been isolated. This protein, which we designate tau (tau), is present in association with tubulin purified from porcine brain by repeated cycles of polymerization. Tau is separated from tubulin by ion exchange chromatography on phosphocellulose. In the absence of tau, tubulin exists entirely as a 6S dimer of two polypeptide chains (al...

PPR8522 encodes a chloroplast-targeted pentatricopeptide repeat protein necessary for maize embryogenesis and vegetative development

The pentatricopeptide repeat (PPR) domain is an RNA binding domain allowing members of the PPR superfamily to participate in post-transcriptional processing of organellar RNA. Loss of PPR8522 from maize (Zea mays) confers an embryo-specific (emb) phenotype. The emb8522 mutation was isolated in an active Mutator (Mu) population and co-segregation analysis revealed that it was tightly linked to a...

Morphogenesis of maize embryos requires ZmPRPL35-1 encoding a plastid ribosomal protein.

In emb (embryo specific) mutants of maize (Zea mays), the two fertilization products have opposite fates: Although the endosperm develops normally, the embryo shows more or less severe aberrations in its development, resulting in nonviable seed. We show here that in mutant emb8516, the development of mutant embryos deviates as soon as the transition stage from that of wild-type siblings. The ba...
